Glucose 6-phosphatase activity was studied histochemically in the oval cells of rat livers during the early stage of hepatocarcinogenesis induced by 3'-methyl-4-dimethylamino azobenzene. Short-period perfusion fixation of the liver preserved both the ultrastructure of the liver cells and glucose

considered the possibility of the transformation of the oval cells into hepatocytes (5, 16, 21, 31), but many others were not in agreement as to the existence of such transformation and rather considered the cells to disappear by cell death and removal (9, 22, 23, 25, 26). Inaoka (1 1), in our laboratory, examining the early stage of azo dye hepatocarcinogenesis in rats, concluded that the oval cells were able to transform into hepatocytes through various transitional stages. This is one of the series of studies (2, 12) to reevaluate this conclusion, investigating the oval cells during azo dye carcinogenesis with application of the electron microscopic histochemical method for examining glucose 6-phosphatase activity which was known to be demonstrable in hepatocytes in normal livers (1).

THE CYTOTOXIC EFFEC T OF CHELIDONIUM ALKALOIDS ON CELLS ISOLATED FROM ADULT RAT LIVER

Alkaloids of the aerial parts of chelidonium were extracted in the salt form, and their aqueous solutions were prepared in different concentrations. Rat hepatocytes were obtained by liver perfusion. The alkaloidal solutions were added to suspensions of hepatocytes in petri dishes and the mixtures were incubated. Two types of controls have been used in one type. no alkaloidal extract was ad...

Isolation of oval cells by centrifugal elutriation and comparison with other cell types purified from normal and preneoplastic livers.

Oval cells and biliary epithelial cells were isolated from livers of rats fed a choline-deficient diet containing 0.1% ethionine and from normal rat livers, respectively. Nonparenchymal cell suspensions prepared from these livers by collagenase perfusion followed by digestion of undissociated tissue with 0.1% collagenase, 0.1% Pronase, and 0.004% DNase I were separated into six fractions by cen...

VARIATIONS OF GLUCOSE 6-PHOSPHATE DEHYDROGENASE ACTIVITY IN VARIOUS TISSUES INDUCED BY METABOLIC ALKALOSIS, ACIDOSIS AND DIABETES

The effects of chronic metabolic acidosis, alkalosis and alloxan-induced ketoacidosis on G6PD activity of rat kidney, liver and erythrocytes were studied. Metabolic acidosis significantly increased the activity of kidney enzyme (55%) but decreased the liver (43%) and erythrocyte (38%) enzyme activities. Alkalosis did not make a significant change in the kidney or liver enzyme activity but ...

The Effect of 6 Week Resistance Training Program on Foxo1 Expression in Liver Cells and Glucose and Insulin Levels in Type 2 Diabetic Rats

Background & objectives: Foxo1 is a transcription factor in insulin signaling in the liver. The function of Foxo1 is important in the ability of insulin to regulate hepatic glucose production and glucose homeostasis. The aim of this study was to determine the effect of resistance training program on Foxo1 expression in liver cells as well as glucose and insulin serum levels in male Wistar rat...

Irreversible fixation of increased level of muscle type aldolase activity appearing in rat liver in the early stage of hepatocarcinogenesis.

azobenzene for 60 days. The muscle type aldolase in the livers of rats thus treated was found to be elevated when compared to nor mal liver, although no appreciable differences were ob served in total aldolase activity, mitotic index, histological architecture, and soluble protein, as well as DNA content. The appearance of this enzymic change was already observable by administrating the azo dye...
